Known as Kaizu Castle built by Takeda Shingen, this was the most critical stronghold during the Battles of Kawanakajima. During the Fourth Battle (1561), it served as the Takeda frontline base. Sanada Nobuyuki entered during the Edo period, and the Sanada governed for 10 generations. Sakuma Shozan, pioneer of opening Japan, was from this domain.

At the Fourth Battle, Kenshin crossed the Chikuma River by night from Mt. Saijo-yama and charged the Takeda main camp at dawn. According to tradition, Kenshin spotted the cooking fires rising from Kaizu Castle and surmised Shingen's strategy, prompting him to move first. The movements of Kaizu Castle proved decisive in the battle's outcome.

Kaizu Castle (modern Matsushiro), laid out by Yamamoto Kansuke around 1560, was Shingen's frontline fortress for the Kawanakajima campaigns. During the Fourth Battle (1561), troops sortied from here to meet Kenshin's dawn assault at Hachiman-hara. It was Shingen's central stronghold for governing northern Shinano.

After Sekigahara, Ieyasu appointed Masayuki's eldest son, Sanada Nobuyuki (Nobushige), as daimyo of Matsushiro Domain with 100,000 koku. After Masayuki's death, Nobuyuki settled in Matsushiro, and the Sanada ruled the domain for ten generations. The Sanada Treasure Museum holds armor and letters associated with both Nobuyuki and Masayuki.
