During the 1582 Battle of Uozu Castle, Uesugi Kagekatsu's forces held out at Matsukura Castle against a fierce siege led by Sassa Narimasa, a vassal of Oda Nobunaga. This site is also historically noted as the place where news of Nobunaga's death in the Honno-ji Incident arrived, a moment that dramatically affected the course of the battle in Etchu.

In 1570, Uesugi Kenshin attacked Matsukura Castle after its lord, Shiina Yasutane, defected to the rival Takeda clan. The castle held out for 100 days, forcing Kenshin to withdraw temporarily, but in 1572 he besieged it again and finally captured it. Thereafter his retainer Kawada Nagachika served as castellan, making Matsukura a key Uesugi stronghold in Etchu.

In 1582, when Oda forces besieged Uozu Castle, Matsukura's castellan Jojo Masashige was dispatched under orders from Uesugi Kagekatsu to relieve it. On June 2, news arrived that Nobunaga had been killed in the Honnoji Incident; Uozu Castle fell the next day, but the Oda army then withdrew. Uesugi forces regrouped at Matsukura Castle and went on to retake Uozu.
